Is there a limit to how many mobs can spawn from a spawner before you activate a farm and kill them all?

You have to take mobs out of escape area for mob spawner to spawn more than 6 mobs.
Escape area is a 16x16x8 cube around the spawner. For more facts about it, google for youtube videos: mob spawner facts. etc.
If spawned mobs are outside this area, then mob spawner will spawn mobs indefinitely, allowing you to get as many mobs as your computer can handle.
I read that on the wiki today. btw this is my 100th post!

The limit for "naturally spawned" (ie. not from a spawner cage, spawn egg, command) hostile mobs is 70 in singleplayer and varies in multiplayer based on the number of players and their geographic distribution [q.v. https://minecraft.gamepedia.com/Spawn#Mob_cap ]

The cap is also dimension specific, eg. 70 zombies in the overworld will not impact the number of Endermen spawning in the End.
An Enderman farm will thus max out at 70 (barring occasional temporary overages) and assuming you are far enough from any End cities that no shulkers count toward the cap.
Other than killing some to drop the number below the cap, the only ways to get more to spawn once you have 70 are spawners (Enderman spawners do not exist in unmodded vanilla), spawn eggs (also not available in unmodded vanilla), commands, and mods.

Also note that there is now something called maxEntityCramming that will suffocate and kill mobs if more than 24 gather on 1 block. That feature was designed to protect servers from runaway mobs farm, but it's on by default in single player as well. /gamerule maxEntityCramming=0 will turn it off. Otherwise you will be limited to 24 mobs, or you'll have to design your collection area to work around that rule (ladders or vines circumvent the rule, or a larger collection area can be built).
Also note that there is now something called maxEntityCramming that will suffocate and kill mobs if more than 24 gather on 1 block. That feature was designed to protect servers from runaway mobs farm, but it's on by default in single player as well. /gamerule maxEntityCramming=0 will turn it off. Otherwise you will be limited to 24 mobs, or you'll have to design your collection area to work around that rule (ladders or vines circumvent the rule, or a larger collection area can be built).
