You can only use those x y z arguments if the command uses x y z arguments. Example: /setblock <x> <y> <z> <TileName> [dataValue] [oldBlockHandling] [dataTag]
........./setblock 4 ~ 12 lava
Do you mean like an xmin and xmax argument? If so, there is a way to do that, with the x, dx, y, dy, z, and dz arguments. Use the x, y, and z arguments to select one corner of a box, then use dx, dy, and dz to select how far in any direction you want the box to extend.
For example, if one corner is on 100,63,42, and you want to test in a 5x5x5 box, the testfor command would be:
testfor @a[x=100,y=63,z=42,dx=5,dy=5,dz=5]
