If I had to guess I'd say that some things that will be added are: A way to get the circular stone bricks in survival (possible now but only with a glitch). I'm also guessing there will be some development around the sandstone wells and the addition of the hieroglyph blocks to the game. They will probably add a few more things to the jungle as well.
However, at this point it's all speculation. This is just my best guess.
Sorry to spoil your fantasy dreams but let's check some facts:
1) Next update will be mainly about the server-side modding API
2) The new mojangstas from Bukkit are not only creating the modding API but are also fixing bugs, especially multiplayer bugs (I can't find the source for that, sorry).
3) Probably a slightly better AI, maybe more complex Villagers' behavior (no source, but I doubt Jon will spend the next weeks doing nothing).
4) As usual, there may be new stuff but nobody really knows.

Probably this. On the video it doesnt says when they are going to do it, but Jeb says he will do it, so it might be for 1.3 or 1.4 who knows
I thought every mod was part of minecraft therefor Mojang was free to implant it into the game at will?
I guess not because of some kind of rights that have the modders over the programming they made. Basicly its mojangs game but its modders mod so they have to make an agreement before anything. I ignore how was the deal with the piston mod but they managed to do it.
Mojang is ****ed up on money, i don't think the optifine team is asking something that mojang can't pay.
From what i heard, its totally backwards, those who are buggin bout money are the optifine guys. I cant blame them really and i dont know much about it so i prefer to leave my statement like this.
The only things we know for a fact are that it will be an update about the Mod API and new AI.
I believe they said that the Mod API will [Or should be] done by 1.3, which is nice.
And Jens said that he wants the player to be able to interact with the villagers and trade with them, which will hopefully come as well.
I would say that an update just about the Mod API would be expected, although considering that it won't be in use at all until it's done unless they want to release a half-finished program, I guess that each week we'll still get bug fixes and tweaks and a few new things.

I agree optimizing the game should be their top priority now. I've seen dozens of posts on the forums citing performance issues.

This is possibly going to be one thing they might do shortly, not sure if they ae going to do it for 1.3.

Totally agree, new additions should be skipped in favour of bug fixes., there are just too many OLD bugs now that need to be fixed.
