I'd like to create an add-on pack for another texture pack which will add some new textures to create a nautical theme. It obviously won't be using any of the original textures, just my own created ones, then you'd layer my pack over the other in minecraft to make a combination. Since I'm not actually modifying or reusing any of their assets, would permission still be required?

If you're not using anything at all from the original pack, not even using the original textures as a base or background, then no you wouldn't need permission. This is exactly the sort of issue that pack stacking is supposed to resolve, so you're doing it right.

If you're worried about that, just make a point to separate your pack from whatever you're making it compatible with. Don't advertise it as an "[name-of-pack-here] add-on" or anything. Branding is also part of the original pack's identity you know. Just have it be a pack unto itself in a similar style for a particular purpose.
That said, if you can contact the original pack's author I STRONGLY suggest doing so. Pack artists frequently LOVE people making add-ons for their work and might even link to your add-on if you publish it. Plus you might get permission to use the original's branding to make yours more identifiable. A little cross-promotion is good for everyone, you know?
Only issue with that is that I'm doing this for Glimmar's fantastic steampunk, but he's been markedly silent for nearly two years... I shall endeavour to contact him all the same.
