Well first you would need a painting program , doesn't really matter what kind, and then just make small 16X16 pictures. Once you are done you put em all together and BAM! you have a texture pack.

Fixed that for you. MSPaint (The default paint program that comes with Windows) does NOT work for textures as it has no transparency support. Instead you want Paint.net (windows only), GIMP or Photoshop (If you have it.)
