Hmm, I wonder how they would react to implementation of martial law/militarization of the village? {Higher, iron walls with iron bars barbed wire on top, occasionally-firing TNT artillery in the town center, iron golem patrols, installation of floodlights, construction of a town hall/fortress, ETC.}
Might I suggest testing their reaction to the domestication of animals? Or perhaps their use of shiny rocks (emeralds to us more civilized folk) as a currency? Why?
I want to test its vision and ability to show any kind of moods - imprison 4 villagers - one in emerald cage, another in golden, third behind iron bars and 4th between TNT and cacti walls. What are their reactions? Also I would like to put them in huge windowless building and wonder will they just jump out of their windows or not if you spawn a zombie?
I love doing stuff like this myself, I don't really do experiments on them, but merely live among them and improve on their villages and try to keep them safe best I can at the same time
Place a large fire pit in an open, populated place of town. This will determine their instincts (whether they run from the fire or flock to it), their intelligence (whether they touch the fire or not), their combustibility and their tolerance for pain!
Let's see how well they can defend themselves when a mysterious cage containing a tiny green man is placed in their town hall! (Zombie spawners using command blocks.) Perhaps they will have to invent some sort of electrical defense mechanism (redstone), construct iron servants (golems), or make traps (lava pits)! Actually, they would probably fall into their own trap.

make a minecart or another means of long distance transportation, and dump a villager in the middle of the wilderness, far from the village.
How will the villager react to this? will it simply walk back to its home? or be lost forever? Will it take the easiest path back to the village? Will it realize what is happening to him as he is being carted away from the village and jump out? Also how will it survive in the new enviroment?
Put a block of TNT next to one of their pressure plate tables, then stuff a whole bunch of the villagers into the house until it is so full they hey to blow each other up to create more room! It's the funniest thing ever as they all look at the one guy who stepped on the plate!

I like to destroy all doors except for in one building, so they all flock to it at night. At the entrance of the building, I put a few trapdoors with pressure plates so when they attempt to leave, they fall into a chamber underground. I then proceed to place them into individual cells, decorated with iron bars and a one-way iron door, with a 1x1 gap window so I may barter with them. If I'm feeling generous, I add a bed to their cell. This way, I can access each of them individually without fear of them running off or getting killed. They also don't seem to mind being locked up.
I also make a point to collect all books from libraries (they certainly don't read, after all), and replace all glass pane windows with wooden fences, to give it that pleasantly eerie, abandoned feel.
Test their reaction to all doors being removed from "Village"
