This is all the information we have currently. I don't know if you will be able to use all mob heads as trophies, or just skulls, but it looks pretty neat.

Dinnerbone then says that "Well there is only one thing, that Jens has been working on awhile that we haven't really talked about." he then says that he will give away a Minecraft sword to someone who guesses the answer to a question he is going to ask.
Dinnerbone: "The new boss that Jens has been working on..."
Audience: "Wither! Witherboss!"
Dinnerbone: That isn't the question by the way."
Everyone has a good chuckle.
Dinnerbone: "It is called the Wither, not the Witherboss, but that's not really official. Um.. Everyone has been asking us, constantly, where does it spawn? Are we going to add a new dimension, are we going to put it in the Nether, are we going to put it in the Overworld or in a new dungeon or something?"
"Who can guess, where or how you spawn the boss?"
There are a flurry of guesses that you can't quite make out in the video. Eventually though, someone says something about skulls and both Jeb and Dinnerbone point at him. They then hand him the sword.
Dinnerbone then asks if Jeb want's to explain it, Jeb suggests that they shouldn't, but the audience member, David, suggests that putting 3 skeleton heads in a row creates the Wither.
Jeb: "There's a little more to it but that's the, uh, that's the basic idea."
Dinnerbone then repeats it for the people at home.
Dinnerbone: "The general idea is that, there's no specific location that you spawn it, but there's a task to spawn it. So you could probably have a thousand Witherbosses, if you work really, really hard for it. Um... We are going to have these really rare items that drop from monsters that are, basically, their heads. Skeleton heads or creepers heads. Um... the idea is that you put these together in a certain, special way, kind of like a ritual. And then, you get punished for it."
Then they joke around a bit and go on to the next question.
So, this confirms that heads will be dropped from monsters, and that it will not just be skeleton heads. And that arranging these heads in a certain way will summon the Wither. You can have as many Wither bosses as you like in a world, and in any dimension.

The snapshot containing mob heads is now out!
The available heads are: Skeleton skull, Wither Skeleton skull, Zombie head, 'human' head and Creeper head.

The heads are also as fully rotatable as signs:

While heads must be placed on a solid block, you can remove the solid block afterwards, much like pumpkins. Although, you can also place them against walls, which does not need a block underneath to be placed and will only face outward.
Also the Wither can be summoned with three Wither skulls on top of soul sand like so:
You must place one of the wither skulls last or the Wither will not be summoned
Wither skulls are from the new Wither skeleton mob, found in the Nether. Use a skeleton spawn egg in the Nether to spawn one. They also seem to drop coal, bones, and partly damaged stone swords, the ones they wield presumably.
You can also wear them on your head! it seems to be a bit glitchy though, displaying over your head but underneath the hat layer, unlike pumpkins. Wearing them does not make mobs non-agressive.

Heads placed in item frames will display the item sprite rather than the 3d head.
Also, heads refused to be pushed or pulled by pistons, much like obsidian, and unlike pumpkins, which break when pushed.
I think that's awesome. Now players can build bases with a real barbaric feel. Not to mention we can now make pyramids out of skulls like certain conquerers use to. It will also be a good decoration for dangerous areas in adventure maps.
On topic though, it's very nice. I look forward to putting Endermen heads above my fireplace.
If it works with all mobs, I look forward to creating a trophy room, featuring the Ender Dragon's head along side the dragon egg (from birth till death...)
Although I wonder how it would work with creatures like the slime. Will we get a mini slime to place? And what about the Wither, do we get all 3 heads? It'll be interesting to see.
Just had a brilliant idea. Ghast head mounted in item frame in front of dispenser that shoots fire charges. I guess it would look better if the ghast head had it's shooting face on (>: O).
OH MY GOD, YES!!! It would be really cool if we could put torches in them, too :3

edit: I think what I might like better than this being a rare drop would be if you could craft it from bones. Yeah, I know, doesn't exactly track. It would be cool if you could give certain villagers items and they could craft them into things like this for you. Like give them 64 bones and they'd give you a skull.
They look to me rather like they have been crafted out of stone , not drops. It will be great if this will be the begining of alowing us to craft items out of stone or "marble".
Since zombies have the ability to wear blocks on their heads, via third party editing, in theory you could get them to wear a skull on their heads. Although since the block is smaller than a normal block it would probably look weird.
If the player has the ability to wear them though they would probably fix any scaling issue. That'd be pretty good.

I think this will be a drop only for skeletons because they're more likely to fall apart.
Maybe you get it if some other mob kills the skeleton(like the way we aquire music discs now, only with skeletons)

I love it!

villagerenderman head in my house.The skulls look about the same size as a skeleton's head, so it might work.
