This looks awesome, I'm imagining towns of people on SMP, with people working together and having different roles, since it's more practical to have 5 people going to different areas of biomes and collecting lots of their specific resource, than each person travelling a lot just to get a single cacti or a bit of sand or some wood. Or even separate mining camps that are in an ore rich biome, that trade with a main city or something. Then there can be shops and an economy, and it'll give a bit more of a teamwork or RPG feel to it, which I'd like. Of course that doesn't have to be the case on every server, but it still allows for it, and I'm all for more options and content to flesh out minecraft.
Bigger Biomes would encourage players on SMP to spread out. Many people who play MC have a biome they prefer. Some people like building pyramids in deserts. Some enjoy playing in igloos in snowy climes. And others like the more neutral forest biomes with their high wood content. Each has it's advantages... and disadvantages. Deserts obviously have lots of sand and cacti, but little water. Snowy climates have Ice (which is not currently mine-able), snow, etc. I propose that these differences be heightened, thus giving players reasons to either travel to far off lands, or trade with those that do.
I also think it would be nice if ores and plants varied by biome. Make cacti only grow in desert biomes (or the 'arid mega-biome'). Make some biomes have different concentrations of some ores. Make wheat not grow in snowy biomes. Stuff like that. New mobs, too. Scorpions- the spiders of the desert. Deer- the cows of the forest. Seals- the pigs of the tundra. Etc.

Each biome should not just look slightly different, but really PLAY different in a notable way. More biome centric flora and fauna would be amazing. Also making it so that certain temperatures/biomes can only grow some plants would make locational farming more interesting. Notch long ago mentioned adding heat and cold damage too from overexposure to harsh biomes, so if that also effected animals it would make ranchs biome centric too.
But really, this is a great idea, and would force players to set up multiple bases.
I doubt we will receive entirely new fauna, but perhaps some of the current fauna will be unavailable in certain biomes, or they will have different textures. Wolves are already a bit like this. Plus, it would be simple to implement your wheat idea. I can think of an equivalent for arid biomes: Wheat cannot grow unless the farmland is hydrated.
Actually, I just realized I messed up my earlier calculations. I somehow did an accidental m-> km tranlation, which made my calcualtion s nonsensical, and I resorted to estimating based on river pixels.
Corrected version:
a chunk is 16x16, or 256 blocks of surface area. 256*70,000 = 17920000 m^2, or a 4233km per side, if it was square. The map is 1765x994, or 1,754,410 pixels. This translates to 10 blocks per pixel, so each pixel is approx a 3x3 area. the image is therefor 5658x3180, approx 5.7kmx3.2km, for a surface atrea of 18.24 km^2
The river is approxamately 20 to 30 blocks wide. the river is approxamately 3 or 4 pixels wide. This gives a scale of roughly 7 blocks per pixel. The image is 1765x994, , which means it is 12.3kmx6.9km, for a surface area of 85 km^2.
the entire island is approximately 3.4 kilometers long then, and crossing 5 biomes gives an approximate size of 680 meters per biome
This estimate is right around the factor of 2 error I had guess from operating off river pixels.
A minecart's max speed is 8m/s. This means it will take approx 85 seconds, or a minute and a half, to cross a biome via minecart. On foot, it is approx double that, or 3 minutes, without accounting for slowdown due to terrain.
Traversing across the entire island by minecart would take 7 minutes, or 14 minutes on foot. Since a minecraft day only lasts 10 minutes, you will not be able to cross the entirety of the island in a single day without braving the night.
Sprinting may reduce these travel times.

I really like the idea of larger biomes but we really need more potenency from our portals. Right now the 1to8 ratio is just not enough. I feel like i'm running a long distance marathon through the nether sometimes just to get to a remote base that is not even a map distance long.
I'm sure it would be of immense use to you people who are drawing conclusions from this picture to note that Notch is still experimenting with all the various numbers to do with Terrain generation of bigger oceans and islands, so the biome image may not even be generated by the same terrain generator as the river screenshot.
Additionaly, doesn't it seems odd that notch, who likes nice, even numbers like 16 and 256, would have such a weird pixel-to-block ratio for this image? I think that sounds a bit strange.
This could very well be a test of terrain gen a bit scaled down from the river screenshot. ALSO, he said blatantly that the generator that made the river image was different from normal, and had amped-up height contrast. If that number in the terrain gen was different, who says none are different here?
