I don't want to do any mini modding here but since you asked, once you submit an application to be a moderator they handle everything. It's hard to explain but they have a thread posting about it.
I am not familliar with this but I think they look for someone who really wants the job. Not for the power, but for the joy of helping other people!
That and it's not all based off of your application, it's like a resume in real life.. You can lie on it (not recommended, at all) or stretch the truth a bit (still not recommended). I usually when I look over applications check who the person is as a member of the minecraftforums community. I look if they understand the rules and try to help report any rule breakers and rebels. Quality posts are key but don't stress every time you post, contribute to the topic or thread and you won't have a problem with 'not quality' posts.
I do have one question, does your reputation count affect this, or the time you have spent on the forum?
Reputation as in (+1's)? No. +1's don't show that you would be good to be a moderator. Most players on the forums that have a high amount of reputation, are people that make funny/spammy post. Some Mods are like that, but stay within the MCF rules. So, no reputation dose not effect your chance.
Applicants that spend a lot of time on the forums might have a higher chance of being accepted, but if that players fools around all the time he/she might not be as good as a player that is only on for 30 minutes. It really depends on how you spend your time on the forums.
Honestly the admins seem to deal with more frustrated people than anything, the mod is just the messenger.
This might be more true than you know. When my mods had someone that was out of hand, I always told them to add me. I was the final word in the conversation with relation to the infraction (whether it was in favor for the moderator or for the member). If the person had additional questions unrelated to their specific infraction that was fine, but my main job after being added to the PM was to make sure that 1) the infraction was fair, 2) the member understood why they were given an infraction, and 3) make sure the member stopped causing any problems for my moderator.

A. Does it make a difference how many times we apply to be a moderator?
B. If we do become a Moderator, can we play the "Catch-up" game if we tell a higher Moderator if we can't get on that week?

C. If we apply for more than 1 forum and get accepted do we get to chose which one we go on, or is it just given to us? (I don't think this one was asked)

Only one Forum Admin gets to pick you, and they have their assigned sections. They choose for you, not the other way around. Though there are certain times when you are given a choice, don't count on it though, and as I think you should only apply for the section you're interested in.
You can however, request to switch to certain sections. Though that is entirely at the discretion of the admin team. But really, why switch when you've applied to the one section you're interested/can dedicate to in?
I totally agree with cestislife but I would like to add that when you have applied 1 or 2 times, you can be sure that your message has been seen and that people have been thinking really hard. If you aren't accepted you probably aren't suitable to be a mod yet so it's no use sending another application straight after the other ones. You might want to wait a few months before sending a new one, instead of spamming us with more applications.
If you do not have timeti read through 2,000 posts then you would not have time to be a moderator in my opinion. Mods are keeping the forums a safe and friendly place for everyone so they have to practically read every thread and post to make sure they are following the rules and a not off topic.

Or will that depend on how long you can be on the site checking for bad threads, post's Etc?
Its only as hard as you want it to be.
Define "Hard" Do you mean time consuming?
Dealing with ticked off people would be kinda hard. Finding the bad content to be removed wouldn't be that hard if you knew the rules.

No, but constantly applying after each batch would make the impression that you're being desperate.
Your reference of 'higher moderator' would be the Forum Admins. Just tell them of your inactivity.
